Setting up FCS Express for Sapphire

Changing the user options for Sapphire Data

1) Select File->Edit Preferences from the main menu. Selecting Edit Preferences

1a) Go to the File Dialog Category.
1b) Check the Allow Multiple Datasets check box. The Dataset Name Keyword edit box will now become enabled.
1c) Enter @DataID in the Dataset Name Keyword edit box.

The screen should now look like the one below.

From now on, when you select an FCS data file to use, and it is a compound Sapphire file, you will be able to browse into the file as if the file was a folder, and select which data set you want.  For instance, the file CBC+CD348.complete , shown below, is a compound Sapphire FCS data file.

If you double click on that file within FCS Express, it will "browse" into that file and show a list of all the data sets that the file contains, as in the picture below.

You can now click on any one of the data sets, and it will open in FCS Express as if it was a regular data file.

2a) Go to the Plots->2D Plots category.
2b) Change the Cells To Show drop down list to display all cells.
2c) Tick on the baseline offset checkbox.

3a) Go to the Plots->1D Plots category
3b) Change the Plot Resolution to "Specified Below"
3c) Change to show 512 resolution

The 32,768 channel data will now be smooth into 512 channel histograms. This makes the populations much easier to visualize.

4) Press the OK button at the bottom of the Edit Preferences screen to save your changes.Cell-Dyn Sapphire is a registered trademark of Abbott Laboratories.
