Why does compensation from my FC 500 listmode files seem incorrect?Coulter listmode data files really contain two datasets inside. The first data set is 10 bit FCS 2.0 data. This is the data set you are used to seeing. The second data set is 20 bit FCS 3.0 data. Typically, users use the first data set, although FCS Express can load the second if the user wishes. The first data set is saved already compensated, there is no way to uncompensate it. The first data set also contains the compensation matrix that was used with the first data set. FCS Express has several options regarding what to when it encounters a data file that has a compensation matrix embedded. This can be found in File->Edit Preferences on the General category as Default Compensation To Use. If you are using the first data set of the Coulter file, set this value to none. If this is not set to none, you are telling FCS Express to apply the compensation matrix to a data file that is already compensated, causing overcompensation. |
